The audio quality would need to be good to have a good translation. You need to be talking with someone else though. Whether you play the audio you want to translate into Skype somehow, or someone else does it on their end, you would need to connect to someone else the whole time.

The microphone option in the Skype translator is set to use the default device but you might be able to get other options to appear somehow since there is a select box. (I don't know how you feed in other audio sources.) I don't have a webcam that works so the only thing I have ever tested was calling with a microphone to the Echo / Sound Test Service to test translation.

It's been in development for the past few years but it might have other options eventually.
